目的探讨阿魏酸钠(sodium ferulate,SF)与苦参素(kushenin)即氧化苦参碱(oxymatrine,OMT)联合用药的协同抗炎作用。方法采用小鼠耳肿胀、大鼠足肿胀、大鼠棉球肉芽肿模型观察SF与OMT联合用药的抗炎作用。体外采用RAW264.7细胞观察联合用药对脂多糖(LPS)诱导的NO合成的影响,用Griess法测定NO生成量。结果 SF与OMT联合应用时,可明显降低二甲苯所致的小鼠耳肿胀和角叉菜胶所致的大鼠足肿胀,大剂量可减轻大鼠棉球肉芽肿的质量,但单独给予SF或OMT对上述3种炎症模型均无显著作用。体外实验结果显示,SF与OMT联合应用可明显降低LPS诱导的RAW264.7细胞NO的产生,且二者之间存在协同作用。结论 SF与OMT联合应用具有明显的协同抗炎作用,降低LPS诱导的炎性因子NO的产生,可能是其减轻炎症反应的机制之一。
Objective To investigate the synergistic anti-inflammatory effect of combination of sodium ferulate (SF) and kushenin (oxymatrine, OMT). Methods The anti-inflammatory effects of combination of SF and OMT were observed in mouse ear swollen, rat foot swollen and rat model of cotton ball granuloma. RAW264.7 cells were used in vitro to observe the effects of the combination therapy on lipopolysaccharide (LPS) -induced NO synthesis, and the NO production was measured by Griess method. Results SF combined with OMT could significantly reduce xylene-induced mouse ear swelling and carrageenan-induced rat foot swelling, high-dose can reduce the quality of cotton ball granuloma, but given SF alone Or OMT had no significant effect on the above 3 kinds of inflammation models. The results of in vitro experiments showed that the combination of SF and OMT could significantly reduce the LPS-induced NO production in RAW264.7 cells, and there was a synergistic effect between them. Conclusion SF combined with OMT has obvious synergistic anti-inflammatory effect and reduces the production of NO induced by LPS, which may be one of the mechanisms to reduce inflammation.
