目的 :观察左肾静脉不全结扎大鼠肾AQP1和AQP2mRNA表达的变化 ,探讨肾静脉压增高引起肾小管重吸收减少的分子机制。方法 :成年雄性Wistar大鼠随机分成左肾静脉不全结扎组 (VCG)和假手术组(SOG) ,用左肾静脉不全结扎法复制精索静脉曲张 ,2月后进行Northernblot分析。结果 :与SOG相比 ,VCG大鼠左肾AQP1mRNA表达强度明显减弱 ,AQP2mRNA表达强度则无明显差异 ,睾丸组织无杂交信号。结论 :左肾静脉不全结扎大鼠肾静脉压增高引起肾小管重吸收减少可能与AQP1mRNA表达减少有关 ,在集合管可能存在激素逃逸的现象
Objective: To observe the changes of renal AQP1 and AQP2 mRNA expression in rats with left renal vein insufficiency (LVEF), and to explore the molecular mechanism of decreased renal tubular reabsorption after increased renal venous pressure. Methods: Adult male Wistar rats were randomly divided into left renal vein occlusion group (VCG) and sham operation group (SOG). Left ventricular insufficiency was used to replicate varicocele. Northern blot was performed after 2 months. Results: Compared with SOG, the expression of AQP1mRNA in left kidney of VCG rats was significantly weakened, but there was no significant difference in the expression of AQP2mRNA between testis and testis. Conclusion: The decrease of renal tubular reabsorption induced by the increase of renal venous pressure in rats with left renal vein insufficiency may be related to the decrease of AQP1mRNA expression, and there may be hormone escaping in the collecting duct
