探讨MS4A12基因在结肠癌发生过程中的表达及其与结肠癌细胞增殖的关系,利用GEO数据库下载含正常结肠、不典型增生结肠腺瘤和结肠癌组织的基因芯片数据GSE37364,对MS4A12基因表达进行标准化后,再对其进行表达差异分析。通过对结肠癌细胞系Lo Vo进行MS4A12基因沉默,以此研究MS4A12基因对结肠癌细胞增殖的影响。MTT法检测细胞增殖,流式细胞术检测细胞周期,荧光定量PCR检测相关基因的表达。MS4A12基因在正常、腺瘤和结肠癌组织中表达依次降低(p<0.001)。MS4A12基因沉默后,细胞增殖较对照明显升高(p<0.05),且S期细胞比例较对照组升高,G1期细胞比例降低(p<0.05)。本研究发现抑制MS4A12基因可以明显促进结肠癌细胞增殖,且在结肠癌发生过程中其表达逐渐降低,这提示MS4A12可能与结肠癌的发生有关并可为结肠癌早期诊断提供依据。
To investigate the expression of MS4A12 gene in colon carcinogenesis and its relationship with the proliferation of colon cancer cells. Gene microarray data GSE37364 containing normal colon, dysplastic colon adenoma and colon cancer were downloaded from GEO database. MS4A12 gene expression After standardization, the difference of expression was analyzed. The effect of MS4A12 gene on the proliferation of colon cancer cells was investigated by silencing MS4A12 gene in Lo Vo cell line. Cell proliferation was detected by MTT assay, cell cycle was detected by flow cytometry, and the expression of related genes was detected by fluorescence quantitative PCR. MS4A12 gene expression decreased in normal, adenoma and colon cancer tissues (p <0.001). After MS4A12 gene silencing, cell proliferation was significantly increased (p <0.05), and the proportion of cells in S phase was higher than that in control group, while the proportion of cells in G1 phase was decreased (p <0.05). This study found that inhibition of MS4A12 gene can significantly promote the proliferation of colon cancer cells, and its expression gradually decreased in the process of colon cancer, suggesting that MS4A12 may be related to the occurrence of colon cancer and provide a basis for the early diagnosis of colon cancer.
