目的:建立表达人癌胚抗原(CEA)的小鼠肝癌细胞模型。方法:用脂质体介导转染法将含人CEA-cDNA的逆转录病毒重组质粒pLXSN-CEA导入小鼠肝癌细胞,利用含G418的选择培养基筛选及放免法测定癌细胞CEA表达量进行鉴定。结果:成功建立表达人CEA的小鼠肝癌细胞模型。结论:利用基因转染技术能将人癌胚抗原cDNA基因导入小鼠肝癌细胞中,并能在选择培养基中生长,且表达人的癌胚抗原。
Objective: To establish a mouse hepatoma cell model expressing human carcinoembryonic antigen (CEA). METHODS: The retroviral recombinant plasmid pLXSN-CEA containing human CEA-cDNA was introduced into mouse hepatocellular carcinoma cells by liposome-mediated transfection method. The expression of CEA in cancer cells was determined by using selection medium containing G418 and radioimmunoassay. Identification. Results: Mouse hepatoma cell model expressing human CEA was successfully established. Conclusion: The gene encoding human carcinoembryonic antigen cDNA can be introduced into mouse hepatocellular carcinoma cells by gene transfection technology, and can be grown in selective media and express human carcinoembryonic antigen.
