建立了过氧化氢(H2O2)诱导人皮肤成纤维细胞损伤及凋亡模型,并研究几种具有抗氧化作用的复方中草药提取物对细胞损伤及凋亡的影响。采用MTT、Hoechst染色及TUNEL等方法检测过氧化氢诱导细胞后以及加入中草药提取物后细胞的存活率、细胞形态以及细胞凋亡的状况。实验结果显示:在浓度为0.8 mmol/L,作用时间6 h,过氧化氢能诱导人皮肤成纤维细胞损伤和凋亡(p<0.05);而抗氧化类中草药复方提取物等物质能够显著提高过氧化氢诱导的细胞存活率的降低,并能明显减少凋亡细胞的数量(p<0.05),表明其对过氧化氢导致的细胞损伤和凋亡作用具有一定的抑制和防护作用。
A hydrogen peroxide (H2O2) -induced injury and apoptosis model of human skin fibroblasts was established. The effects of several Chinese herbal extracts with antioxidant on cell injury and apoptosis were also studied. Cell viability, cell morphology and apoptosis were detected by MTT assay, Hoechst staining and TUNEL assay after the cells were induced by hydrogen peroxide. The results showed that hydrogen peroxide could induce the injury and apoptosis of human skin fibroblasts (p <0.05) at the concentration of 0.8 mmol / L and the action time of 6 h, while the anti-oxidant Chinese herbal compound extract and other substances could be significantly increased Hydrogen peroxide induced a decrease in cell viability and a significant decrease in the number of apoptotic cells (p <0.05), indicating that it has some inhibitory and protective effects on hydrogen peroxide-induced cell injury and apoptosis.
