目的探讨食管癌瘤内注射生物可降解5-氟尿嘧啶聚乳酸纳米微粒(5-Fu-NPs)缓释剂所产生的抗肿瘤活性,为临床应用提供实验依据。方法36只SCID小鼠随机分为6组,分别是瘤内注射生理盐水的对照组、瘤内注射5-Fu-NPs(包括低剂量和高剂量)组、瘤内注射无载药NPs组、瘤内注射5-Fu组及腹腔内注射5-Fu组。观察荷瘤鼠肿瘤生长,荷瘤鼠给药前、后血象及给药后肿瘤组织的凋亡指数。结果瘤内注射5-Fu-NPs缓释剂组小鼠(20mg/kg体重,q3d×3)肿瘤生长缓慢,瘤体明显小于5-Fu腹腔注射给药组,并呈现良好的量效关系。结论瘤内注射5-Fu-NPs缓释剂的抗肿瘤效果优于局部和全身单纯5-Fu给药,它具有较重要研发价值。
Objective To investigate the antitumor activity of 5-fluorouracil-loaded polylactic acid nanoparticles (5-Fu-NPs) injected into esophageal carcinoma and to provide experimental evidence for its clinical application. Methods Thirty-six SCID mice were randomly divided into 6 groups: control group injected intratumoral injection of NS, intratumoral injection of 5-Fu-NPs (including low dose and high dose), intratumoral injection of drug-free NPs group, Intratumor injection of 5-Fu group and intraperitoneal injection of 5-Fu group. The tumor growth of the tumor-bearing mice was observed, and the apoptosis index of the tumor tissues before and after the administration of the tumor-bearing mice and the blood samples after the administration were observed. Results Tumor growth was slow in mice injected with 5-Fu-NPs sustained-release agent (20mg / kg body weight, q3d × 3), which was significantly smaller than that of 5-Fu intraperitoneal injection. Conclusion The antitumor effect of intratumoral injection of 5-Fu-NPs sustained-release agent is superior to local and systemic administration of 5-Fu alone. It has important value in research and development.
