目的:研究可溶性Jagged1/Fc对DC-T细胞共培养体系中TGF-β1、IL-10、IL-4、IL-2、IFN-γ及TNF-α表达水平的影响。方法:应用IL-4和GM-CSF诱导培养小鼠骨髓细胞,流式细胞术检测髓系树突状细胞(DC)分化标志CD11c;将等量的淋巴细胞与之共培养,ELISA检测共培养上清TGF-β1的含量,Luminex100检测IL-10、IL-4、IL-2、IFN-γ和TNF-α的含量。结果:可溶性Jagged1/Fc处理组TGF-β1和IL-10的水平与DAPT对照组之间无统计学意义(P>0.05),而IL-4、IL-2、IFN-γ和TNF-α的水平升高(P<0.05)。结论:Jagged1/Fc能促进DC-T细胞相互作用导致IL-4、IL-2、IFN-γ和TNF-α的水平上调。
Objective: To investigate the effect of soluble Jagged1 / Fc on the expression of TGF-β1, IL-10, IL-4, IL-2, IFN-γ and TNF-α in DC-T cell coculture system. Conclusion: Jagged1 / Fc can promote the interaction of DC-T cells leading to the upregulation of IL-4, IL-2, IFN-γ and TNF-α.
