目的:研究消臌软坚丸对肝硬化大鼠模型血清肿瘤坏死因子(TNF)、白细胞介素1、6(IL-1、IL-6)、一氧化氮及其合酶(NO、NOS)及血浆内皮素1(ET-1)的调控作用。方法:采用四氯化碳为主的复合因素造模法复制肝硬化大鼠模型,分为正常组、模型组、消臌软坚丸低剂量组和高剂量组及阳性药对照组(复方鳖甲软肝片),5组均于造模结束后第2天连续灌服0.85%氯化钠液或药物28d后断头取血,分别检测血清TNF、IL-1、IL-6、NO、NOS及ET-1含量。结果:①消臌软坚丸各治疗组TNF、IL-1、IL-6的含量均低于模型组(P<0.01),高剂量组低于对照组(P<0.05或<0.01);低剂量组IL-1、TNF的含量低于对照组(P<0.01),而IL-6的含量与对照组之间差异无统计学意义(P>0.05);高、低剂量组之间差异未见统计学意义(I’>0.05)。②消臌软坚丸各治疗组NO、NOS的含量均低于模型组(P<0.01);高剂量组NO、NOS的含量均低于低剂量组和对照组(P<0.01);低剂量组NOS的含量低于对照组(P<0.01)。③消臌软坚丸各治疗组均能明显降低血浆ET-1的含量(P<0.01)。高、低剂量组ET-1的含量均低于对照组(P<0.01)。高剂量组ET-1的含量低于低剂量组(P<0.01)。结论:①消臌软坚丸可显著下调肝硬化模型大鼠血清异常升高的TNF、IL-1、IL-6的水平;②消臌软坚丸可显著降低肝硬化大鼠血NO?
OBJECTIVE: To study the effects of Xiaoyu Ruanjian Pills on serum tumor necrosis factor (TNF), interleukin 1, 6 (IL-1, IL-6), nitric oxide and its synthase (NO, NOS) in liver cirrhosis rat models. The regulation of plasma endothelin-1 (ET-1). METHODS: Cirrhosis rat models were duplicated using a carbon tetrachloride-based compound factor model and divided into normal group, model group, low dose group and high dose group of Xiaoyu Ruanjian Pill, and positive drug control group (Compound armored Soft liver slices), 5 groups were fed with 0.85% sodium chloride solution or drugs for 28 days after the completion of the model, and were decapitated and blood samples were taken. The serum levels of TNF, IL-1, IL-6, NO and NOS were measured. And ET-1 content. Results: 1 The content of TNF, IL-1 and IL-6 in Xiaozhi Ruanjian pill group was lower than that in model group (P<0.01). The high dose group was lower than the control group (P<0.05 or <0.01). The levels of IL-1 and TNF in the group were lower than those in the control group (P<0.01), but there was no significant difference between the levels of IL-6 and the control group (P>0.05). There was no difference between the high and low dose groups. Statistical significance (I’>0.05). 2 The content of NO and NOS in the treatment group was lower than that in the model group (P<0.01). The content of NO and NOS in the high-dose group was lower than that in the low-dose group and the control group (P<0.01); The content of NOS was lower than that of the control group (P<0.01). 3 Xiaotan Ruanjian pill can significantly reduce plasma ET-1 content in each treatment group (P<0.01). The contents of ET-1 in the high and low dose groups were lower than those in the control group (P<0.01). The content of ET-1 in the high-dose group was lower than that in the low-dose group (P<0.01). Conclusion: Yixiao Ruanjian Pills can significantly down-regulate the serum levels of TNF, IL-1 and IL-6 in cirrhosis model rats. 2 Xiaoyu Ruanjian Pills can significantly reduce NO in blood of cirrhotic rats.
