目的建立紫丁香叶中咖啡酸的含量测定方法。方法采用RP-HPLC法测定紫丁香叶中咖啡酸的含量,色谱条件为:DiamonsilTMC18(150mm×4.6mm,5μm)分析柱,流动相:甲醇-0.1%磷酸水溶液(15∶85),检测波长:323nm,体积流量:1.0ml/min,柱温:25℃。结果咖啡酸的进样量在0.0378~0.2268μg范围内与峰面积呈良好的线性关系(r=0.9996)。平均回收率为98.4%,RSD为2.2%(n=5);3批样品中咖啡酸的含量分别为0.01047、0.01103、0.01088mg/g,RSD分别为2.0%、2.2%、1.6%。结论本方法精密度高,分离度好,可用于紫丁香叶中咖啡酸的质量控制。
Objective To establish a method for determination of caffeic acid in lilac leaves. Methods The content of caffeic acid was determined by RP-HPLC. The chromatographic conditions were: DiamonsilTM C18(150mm×4.6mm, 5μm) analytical column, mobile phase: methanol-0.1% phosphoric acid aqueous solution (15∶85), detection wavelength: 323 nm, volume flow: 1.0 ml/min, column temperature: 25°C. Results There was a good linear relationship between the amount of caffeic acid in the range of 0.0378-0.2268 μg and the peak area (r=0.9996). The average recovery was 98.4% and the RSD was 2.2% (n=5). The caffeic acid content of the three batches of samples was 0.01047, 0.01103, and 0.01088 mg/g, respectively, and the RSD was 2.0%, 2.2%, and 1.6%, respectively. Conclusion The method has high precision and good resolution and can be used for the quality control of caffeic acid in the lilac leaves.
