目的研究62例脑胶质瘤细胞中环氧化酶(COX)2、血管内皮生长因子(VEGF)和第八因子相关抗原(ⅧRAg)的表达,探讨COX2在脑胶质瘤中的作用。方法运用免疫组织化学方法研究COX2、VEGF和Ⅷ因子相关抗原在脑胶质瘤中的表达。结果COX2在脑胶质瘤细胞中的胞浆染色阳性,和胶质瘤的病理级别正相关(r=0.874,P<0.01);在坏死区域周围有COX2高表达的肿瘤细胞积聚;肿瘤组织中的血管周围有COX2染色阳性的肿瘤细胞聚集。脑胶质瘤中COX2和VEGF的表达(r=0.4578),COX2的表达和MVD之间(r=0.3918)呈正相关性,差异有统计学意义(P<0.01)。结论COX2的表达和脑胶质瘤的病理分级成正相关,和脑胶质瘤的发生和发展有关系;COX2的表达和VEGF成正相关,COX2和微血管密度成正相关,COX2的表达可促进肿瘤组织新生血管的生成;COX2可作为判断脑胶质瘤恶性度的辅助生物学标志。
Objective To investigate the expression of cyclooxygenase 2 (COX - 2), vascular endothelial growth factor (VEGF) and factor VIII related antigen (ⅧRAg) in 62 glioma cells and to explore the role of COX2 in glioma. Methods The expression of COX2, VEGF and factor Ⅷ related antigens in gliomas was studied by immunohistochemistry. Results COX2 staining in cytoplasm of glioma cells was positively correlated with the pathological grade of glioma (r = 0.874, P <0.01). COX2 overexpression was found around the necrotic area. In the tumor tissues Of the blood vessels around the COX2 staining-positive tumor cells gathered. The expression of COX2 and VEGF in glioma (r = 0.4578), COX2 expression and MVD (r = 0.3918) were positively correlated, the difference was statistically significant (P <0.01). Conclusions The expression of COX2 is positively correlated with the pathological grade of glioma and is related to the occurrence and development of glioma. The expression of COX2 is positively correlated with VEGF, COX2 and microvessel density are positively correlated, and the expression of COX2 promotes the growth of tumor tissue Angiogenesis; COX2 can be used as a secondary biological marker to judge the malignancy of glioma.
