目的:探讨Caspase 3在土荆皮酸(PAB)诱导卵巢癌SKOV3细胞凋亡中的作用。方法:使用不同浓度的PAB处理SKOV3细胞,应用流式细胞仪检测细胞周期的分布,采用Western Blot法检测SKOV3细胞经PAB作用前后Bcl-2和Caspase 3蛋白的表达变化。结果:经PAB作用后,SKOV3细胞出现剂量依赖性G2/M期阻滞伴随细胞凋亡,随药物浓度增加,Bcl-2蛋白表达逐渐下降,Caspase 3蛋白表达上调,Caspase 3活化百分率增高。结论:PAB能通过调节Bcl-2蛋白的表达激活Caspase 3,从而诱导SKOV3细胞发生凋亡。
AIM: To investigate the role of caspase 3 in the apoptosis of ovarian cancer SKOV3 cells induced by germacic acid (PAB). Methods: SKOV3 cells were treated with different concentrations of PAB. The distribution of cell cycle was detected by flow cytometry. The expression of Bcl-2 and Caspase 3 protein in SKOV3 cells was detected by Western Blot. Results: After PAB treatment, G2 / M phase arrest of SKOV3 cells was accompanied with apoptosis. With the increase of drug concentration, the expression of Bcl-2 protein decreased gradually, the expression of Caspase 3 protein increased and the percentage of activation of Caspase 3 increased. Conclusion: PAB can activate Caspase 3 by regulating the expression of Bcl-2 protein and induce the apoptosis of SKOV3 cells.
