目的:研究苦杏仁苷对大鼠肝星状细胞HSC-T6增殖的影响和对凋亡基因Bax的调控作用。方法:利用噻唑蓝染色法动态检测不同浓度的苦杏仁苷对大鼠HSC-T6细胞增殖的影响,利用PCR法检测苦杏仁苷对HSC-T6细胞凋亡基因Bax的表达水平。结果:苦杏仁各实验浓度组对HSC-T6细胞的增殖无直接抑制作用,但其高浓度10-3mol/L和中浓度10-4mol/L可以使HSC-T6细胞提前到达增殖高峰期;而各浓度组对HSC-T6细胞的Bax基因都有一定的诱导作用,并且存在量效关系。结论:苦杏仁苷不能直接抑制HSC-T6细胞的增殖,但可能通过诱导或上调凋亡基因Bax的过度表达,使细胞提前进入凋亡程序阶段,最终使活化的HSC-T6发生细胞凋亡,以起到抗肝纤维化的作用。
Objective: To study the effect of amygdalin on the proliferation of rat hepatic stellate cells HSC-T6 and the regulation of apoptosis gene Bax. Methods: The effect of different concentrations of amygdalin on the proliferation of HSC-T6 cells was detected by thiazolyl blue staining. The expression of apoptosis-related gene Bax in HSC-T6 cells was detected by PCR. Results: There was no direct inhibitory effect on the proliferation of HSC-T6 cells in each concentration of bitter almonds, however, HSC-T6 cells reached the peak of proliferation in advance when the concentration of 10-3mol / L and the concentration of 10-4mol / L were high; The concentration of Bax gene in HSC-T6 cells have some induction, and there is a dose-response relationship. CONCLUSION: Amygdalin can not directly inhibit the proliferation of HSC-T6 cells, but may induce apoptosis in the HSC-T6 cells by inducing or up-regulating the overexpression of Bax, To play a role in anti-liver fibrosis.
