目的观察雷公藤多甙对白细胞介素(IL)-1β诱导大鼠滑膜细胞株 RSC-364基质金属蛋白酶-3(MMP3)、c-Jun mRNA 表达的影响。方法选择不同浓度的雷公藤多甙作用 IL-1β刺激的大鼠滑膜细胞株 RSC-364,37℃,5%CO_2的环境下培养48 h,运用酶联免疫吸附试验(ELISA)检测细胞上清的 MMP3和实时聚合酶链反应(real-time PCR)检测分析细胞中的 c-Jun mRNA 表达。结果不同浓度的雷公藤多甙(0、5、10、20 mg/L)对 IL-1β刺激的大鼠滑膜细胞株 RSC-364分泌MMP3的水平依次是198、176、140、115μg/L,c-Jun mRNA 表达分别是1.17×10~6、3.31×10~5、9.32×10~4、2.81×10~4 copies/ml,即呈现明显地抑制。重要的是:c-Jun mRNA 表达抑制呈现明显的剂量依赖性。结论雷公藤多甙治疗类风湿性关节炎的有效的作用机制,可能与其抑制 MMP3、c-Jun 基因的表达相关。
Objective To observe the effect of Tripterygium wilfordii on the expression of matrix metalloproteinase-3 (MMP3) and c-Jun mRNA in synovial cell line RSC-364 induced by interleukin (IL)-1β. Methods The rat synoviocytes strain RSC-364 stimulated with different concentrations of Tripterygium wilfordii against IL-1β was cultured for 48 h at 37 °C and 5% CO 2 . The cells were detected by enzyme-linked immunosorbent assay (ELISA). Clear MMP3 and real-time PCR assays analyzed c-Jun mRNA expression in cells. Results The levels of MMP3 secreted by IL-1β-stimulated rat synovial cell line RSC-364 in different concentrations of Tripterygium wilfordii (0, 5, 10, 20 mg/L) were 198, 176, 140, and 115 μg/L, respectively. The expression of c-Jun mRNA was 1.17×10~6, 3.31×10~5, 9.32×10~4, and 2.81×10~4 copies/ml, respectively. Importantly, c-Jun mRNA expression was inhibited in a dose-dependent manner. Conclusion The effective mechanism of Tripterygium wilfordii for treating rheumatoid arthritis may be related to its inhibition of the expression of MMP3 and c-Jun genes.
