本文比较了心房钠尿肽 (ANP)、C 型钠尿肽 (CNP)、血管钠肽 (VNP)抑制肺动脉平滑肌细胞 (PASMCs)增殖的效应。用蛋白激酶C激动剂佛波酯 (PMA)刺激体外培养大鼠PASMCs的增殖 ,以总蛋白含量和MTT比色OD值为指标 ,观察三种钠尿肽对PMA刺激大鼠PASMCs增殖的影响。结果表明 ,PMA (10 -9~ 10 -7mol/L)显著升高(P <0 0 5 )PASMCs的总蛋白含量和MTTOD值 ,VNP (10 -8~ 10 -6mol/L)、ANP和CNP (10 -7~ 10 -6mol/L)均可显著降低 (P <0 0 5 )PMA (10 -8mol/L)刺激PASMCs的总蛋白含量和MTTOD值。结果提示 :PMA对大鼠肺动脉平滑肌细胞有促增殖作用 ,ANP、CNP、VNP均可抑制PMA刺激大鼠肺动脉平滑肌细胞的增殖 ,VNP的抑制效应最强。
This article compared the effects of atrial natriuretic peptide (ANP), C-type natriuretic peptide (CNP), and vascular natriuretic peptide (VNP) on the proliferation of pulmonary artery smooth muscle cells (PASMCs). Proliferation of PASMCs cultured in vitro was stimulated by the protein kinase C agonist phorbol ester (PMA). The total protein content and MTT colorimetric OD values were used as indexes to observe the effects of three kinds of natriuretic peptides on the proliferation of PASMCs stimulated by PMA. The results showed that the total protein content and MTTOD value of PMA (10 -8 ~ 10 -6 mol / L), ANP and CNP were significantly increased (P <0.05) (10 -7 ~ 10 -6 mol / L) significantly decreased the total protein and MTTOD of PASMCs stimulated by PMA (10 -8 mol / L). The results suggest that PMA can promote rat pulmonary artery smooth muscle cells proliferation, ANP, CNP, VNP can inhibit PMA-induced proliferation of rat pulmonary artery smooth muscle cells, VNP strongest inhibitory effect.
