[目的]观察血小板型12-脂氧合酶(p12-LOX)在胃癌SGC-7901细胞中的表达及黄芩素(Baicalein,Bai)对胃癌SGC-7901细胞血管内皮生长因子(vascular endothelial growth factor,VEGF)和肝细胞生长因子((hepatocyte growth fact,HGF)表达的影响,并初步探讨作用机制。[方法]RT-PCR检测SGC-7901细胞中p12-LOXmRNA的表达及Bai对其VEGF mRNA表达的影响;;ELISA法检测Bai对VEGF和HGF蛋白表达的影响。[结果]RT-PCR产物经琼脂糖电泳结果,可见一条337bp的p12-LOX基因扩增带和一条651bp的β-actin内参基因扩增带;Bai作用6h后VEGFmRNA表达明显低于对照组(除5μmol/L组),且随Bai浓度的增加,VEGF mRNA表达量逐渐降低;ELISA显示,不同浓度Bai作用SGC-7901细胞24h后,各实验组与对照组相比,HGF蛋白表达量均明显降低(除5μmol/L组),且与浓度相关;随着Bai浓度的增加,各实验组HGF表达量均明显下降。[结论]SGC-7901细胞中存在p12-LOX的表达;Bai可抑制SGC-7901细胞VEGF和HGF的表达,且呈一定的浓度依赖性。
[Objective] To observe the expression of platelet type 12-lipoxygenase (p12-LOX) in gastric cancer cell line SGC-7901 and the effects of baicalein (Bai) on the expression of vascular endothelial growth factor VEGF and hepatocyte growth factor (HGF) in SGC-7901 cells and to explore the possible mechanism. [Methods] The expression of p12-LOX mRNA in SGC-7901 cells was detected by RT-PCR and the expression of VEGF ; ELISA method was used to detect the effect of Bai on the expression of VEGF and HGF protein. [Results] The results of agarose gel electrophoresis showed that a 337bp p12-LOX gene amplification band and a 651bp β-actin gene amplification After treated with Bai for 6 hours, the expression of VEGF mRNA was significantly lower than that of the control group (except for 5 μmol / L group), and the expression of VEGF mRNA gradually decreased with the increase of Bai concentration. ELISA showed that after treated with different concentrations of Bai for 24 hours, Compared with the control group, the expression of HGF protein in experimental group was significantly decreased (except for 5μmol / L group), and correlated with the concentration; with the increase of Bai concentration, the expression of HGF in each experimental group decreased significantly. [Conclusion] SGC -7901 cells exist p12-LOX expression; Bai can inhibit SGC-7901 cells V EGF and HGF expression, and a certain concentration-dependent manner.
