目的研究辣椒素对~(131)I内放射损伤7天后大鼠腮腺的作用及其对烟酰胺磷酸核糖转移酶(nicotinamide phosphoribosyltransferase,NAMPT)表达的影响。方法 Wistar大鼠36只,随机分4个组:对照组、单纯辣椒素组(腮腺区皮肤表面涂抹0.1%辣椒素乳膏,2次/日),单纯~(131)I组(腹腔注射~(131)I,0.5m Ci/100 g),辣椒素治疗组(~(131)I腹腔注射后局部涂抹辣椒素)。给予~(131)I 7天后取腮腺,HE染色法观察~(131)I处理后大鼠腮腺组织的形态学变化,免疫组织化学染色法观察辣椒素对~(131)I处理后大鼠腮腺中NAMPT表达的影响。结果单纯~(131)I组腺泡细胞皱缩明显,核固缩;辣椒素治疗组的细胞形态改变较单纯~(131)I组减轻,未见明显核固缩;单纯辣椒素组与对照组无明显差异。IHC结果显示单纯~(131)I组NAMPT表达量较空白组减少了29.19%(P<0.05);辣椒素治疗组NAMPT表达量较单纯~(131)I组增加了26.29%(P<0.05);辣椒素治疗组中NAMPT蛋白质的表达量与空白对照组和单纯辣椒素组相比均无显著性差异;单纯辣椒素组中NAMPT蛋白质的表达量较空白对照组无显著性差异。结论辣椒素能够减轻~(131)I所致的大鼠腮腺组织损伤,其机制可能与NAMPT表达上调相关。本研究有望为防治~(131)I内放射性涎腺炎提供新的策略。
Objective To investigate the effect of capsaicin on the parotid gland in rats after 7 days radiation injury and its effect on the expression of nicotinamide phosphoribosyltransferase (NAMPT). Methods Thirty - six Wistar rats were randomly divided into four groups: control group, simple capsaicin group (0.1% capsaicin cream on the skin of parotid gland area, twice daily), 131I group (ip) (131) I, 0.5 m Ci / 100 g), and capsaicin-treated group (~ (131) I). Parotid glands were harvested after 131I administration for 131 days. HE staining was used to observe the morphological changes of the parotid gland in ~ (131) I-treated rats. Immunohistochemical staining was used to observe the effect of capsaicin on parotid gland In NAMPT expression. Results The acinar cells in ~ (131) I group had obvious shrinkage and nuclear pyknosis. The morphological changes of capsaicin-treated group were less than those in 131I group, and no obvious nuclear pyknosis was observed. Compared with the control group No significant difference between groups. The results of IHC showed that NAMPT expression in group 131I was reduced by 29.19% (P <0.05) compared with blank group; NAMPT expression in group treated with capsaicin was increased by 26.29% (P <0.05) ; NAMPT protein expression in capsaicin treatment group compared with the blank control group and simple capsaicin group no significant difference; simple capsaicin group NAMPT protein expression compared with the blank control group no significant difference. Conclusion Capsaicin can reduce parotid tissue injury induced by ~ (131) I in rats, which may be related to the up-regulation of NAMPT. This study is expected to provide a new strategy for the prevention and treatment of 131I intraosseous sialositis.
