目的:观察甘油三酯对体外培养大鼠胰岛细胞分泌功能及对细胞内钙离子平均荧光强度的影响。方法:体外分离SD大鼠胰岛细胞进行单层细胞培养,与不同浓度甘油三酯(0、0.25、2.5、5.0、10.0mmol/L)共同孵育72小时,进行葡萄糖负荷试验,放射免疫法检测培养液中胰岛素水平,以评价胰岛细胞功能变化;采用Flou-3/AM荧光染色法,共聚焦显微镜下观察细胞内钙离子荧光强度的强弱,以间接反映细胞内游离钙的水平。结果:5.0、10.0mmol/L甘油三酯可抑制胰岛细胞的高糖刺激下的胰岛素分泌(p<0.05);甘油三酯组(2.5、5.0、10.0mmol/l)细胞内钙离子平均荧光强度均明显增强,与对照组比较有显著性差异(p<0.05)。结论:甘油三酯可以导致胰岛细胞内游离钙水平异常升高,影响细胞的分泌功能。
OBJECTIVE: To observe the effect of triglyceride on the secretion of pancreatic islet cells in vitro and the average fluorescence intensity of intracellular calcium. METHODS: SD rat pancreatic islet cells were isolated and cultured in vitro. The cells were incubated with different concentrations of triglyceride (0, 0.25, 2.5, 5.0 and 10.0 mmol / L) for 72 hours. Glucose load test and radioimmunoassay Liquid insulin level to evaluate the functional changes of islet cells; using Flou-3 / AM fluorescence staining, the intensity of intracellular calcium fluorescence intensity was observed under confocal microscope to indirectly reflect the level of intracellular free calcium. Results: 5.0, 10.0mmol / L triglyceride could inhibit the insulin secretion under high glucose (p <0.05) of islet cells; mean intracellular calcium fluorescence intensity of triglyceride group (2.5, 5.0, 10.0mmol / Were significantly increased compared with the control group were significantly different (p <0.05). CONCLUSION: Triglycerides can cause an abnormal increase of intracellular free calcium in islet cells and affect the secretion function of cells.
