目的:观察陡脉冲电场对荷瘤裸鼠肿瘤生长的影响,分析其杀伤肿瘤细胞的作用机制。方法:建立卵巢癌荷瘤裸鼠模型,设对照组和陡脉冲治疗组,记录治疗后21天各荷瘤裸鼠肿瘤的体积和重量;光、电镜观察陡脉冲对肿瘤组织的病理学改变。结果:陡脉冲治疗组裸鼠肿瘤体积与重量明显低于对照组(P<0.01);陡脉冲对肿瘤组织有明显的破坏作用,治疗靶区内见广泛的凝固性坏死、血栓形成,电镜下出现典型的细胞凋亡改变。结论:陡脉冲通过对肿瘤细胞的杀伤、诱导细胞凋亡和血栓形成抑制肿瘤生长。
OBJECTIVE: To observe the effect of steep pulsed electric field on tumor growth in nude mice and to analyze its mechanism of killing tumor cells. Methods: The ovarian cancer-bearing nude mice model was established, and the control group and steep pulse treatment group were established. The tumor volume and weight of each tumor-bearing nude mice were recorded 21 days after treatment. Pathological changes of the tumor tissue were observed by light and electron microscopy. Results: The volume and weight of the tumor in the steep pulse group were significantly lower than those in the control group (P <0.01). The steep pulse had a significant destructive effect on the tumor tissue and extensive coagulation necrosis and thrombosis in the target area. A typical apoptotic changes occur. Conclusions: Steep pulses inhibit tumor growth by killing tumor cells, inducing apoptosis and thrombosis.
