目的 :初步探讨人参皂甙Rg1对积水肾小管及间质细胞凋亡与Bcl- 2蛋白表达的作用 .方法 :建立大鼠单侧输尿管梗阻模型 ,并给予人参皂甙Rg1灌胃 ,于模型建立后 1周处死大鼠 .除常规光镜检查外 ,免疫组织化学染色法检查Bcl- 2蛋白表达水平 ,末端转移酶介导的dUTP缺口末端标记法检测凋亡细胞 .用病理分析软件分别对肾小管及间质Bcl- 2蛋白表达及细胞凋亡作定量分析 .结果 :单侧输尿管梗阻可导致肾小管和间质细胞凋亡和Bcl- 2蛋白表达增多 ,人参皂甙Rg1显著抑制了输尿管梗阻导致的肾小管及间质细胞凋亡的增加 ,同时进一步促进了梗阻肾小管细胞Bcl- 2蛋白的表达 ,但却抑制了梗阻肾间质细胞Bcl- 2蛋白表达 .结论 :人参皂甙Rg1可抑制单侧输尿管梗阻引起的肾细胞的凋亡 ,其作用机制可能与Rg1调节了Bcl -2蛋白的表达有关
OBJECTIVE: To investigate the effect of ginsenoside Rg1 on the apoptosis of tubular and interstitial cells and the expression of Bcl-2 in hydronephrosis.Methods: The model of unilateral ureteral obstruction in rats was established and ginsenoside Rg1 was administered intragastrically. After establishment of the model The rats were sacrificed at 1 week.Expression of Bcl-2 protein was detected by immunohistochemical staining and apoptotic cells were detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assay.At the same time, And mesenchymal Bcl-2 protein expression and apoptosis were analyzed quantitatively.Results: Unilateral ureteral obstruction caused tubular and interstitial cell apoptosis and Bcl-2 protein expression increased, ginsenoside Rg1 significantly inhibited ureteral obstruction caused Tubular cells and interstitial cells, and further promote the expression of Bcl-2 protein in obstructive renal tubular cells, but inhibit the expression of Bcl-2 protein in obstructed renal interstitial cells.Conclusion: Ginsenoside Rg1 can inhibit unilateral The mechanism of renal cell apoptosis induced by ureteral obstruction may be related to the regulation of Bcl-2 expression by Rg1
