目的 :研究和胃消痞胶囊对大鼠胃肠肌间神经丛一氧化氮合酶 (NOS)的影响 ,探讨其作用机制。方法 :采用隔日禁食的方法制作大鼠胃肠运动功能紊乱模型 ,以辅酶 黄递酶组化染色法及消化道铺片技术显示大鼠胃及上段小肠纵肌层 NOS阳性神经。结果 :胃窦 NOS阳性神经元灰度值模型组 (模型组 )低于正常对照组 ;和胃消痞胶囊大、小剂量组低于对照组 ,但高于模型组 ;中剂量组高于模型组及对照组 ;吗丁啉组低于模型组及对照组。胃底模型组及和胃消痞胶囊大、中剂量组均低于对照组 ;小剂量组及吗丁啉组高于对照组。上段模型组及和胃消痞胶囊小剂量组与对照组接近 ,大、中剂量组及吗丁啉组显著高于对照组。结论 :和胃消痞胶囊可预防模型鼠胃窦动力紊乱 ;大剂量增加胃底 NO合成 ,小剂量使胃底 NO合成减少 ,对胃底容受性舒张功能有双向调节作用 ;大、中剂量可减少上段小肠 NO合成 ,增强小肠蠕动功能
Objective: To study the effect of Weixiao Capsule on the expression of nitric oxide synthase (NOS) in the gastrointestinal myenteric plexus in rats and to explore its mechanism of action. METHODS: The gastrointestinal motility dysfunction model was established by fasting every other day. NOS-positive nerves were found in the stomach and upper small intestine longitudinal muscle layers of rats by coenzyme diaphorase staining and digestive tract preparation. Results: The NS positive neurons gray value model group (model group) was lower than the normal control group; and Weixiaoyu capsule large and small dose group was lower than the control group, but higher than the model group; the middle dose group was higher than the model Group and control group; motilium group was lower than model group and control group. The gastric fundus model group and Weixiaoxiao capsule were lower than the control group in the large and medium dose groups; the low dose group and motilium group were higher than the control group. The upper model group and Weixiaoxiao capsule small dose group were close to the control group, while the large, middle dose group and motilium group were significantly higher than the control group. Conclusion: Heweixiaotan capsule can prevent motility disorders of gastric antrum in model rats, increase NO synthesis in gastric fundus at large doses, decrease NO synthesis in gastric fundus at low doses, and regulate biliary diastolic function in gastric fundus; large and medium doses Can reduce the upper intestine NO synthesis, enhance small intestine peristalsis
