目的利用蛋白质组相关技术分析孕早期胚胎绒毛和蜕膜组织蛋白质组的差异表达,探讨胚胎植入的可能机制。方法选择2007年1-5月期间,正常妊娠10周在本院行人工流产者的胚胎绒毛和子宫蜕膜组织各20例。用固相pH梯度双向凝胶电泳(2D-PAGE)分别分离正常早孕人工流产者的胚胎绒毛和蜕膜的蛋白质组。考马斯亮蓝显色,PDQuest2DE软件分析。结果图像分析测得绒毛与蜕膜凝胶的匹配率蜕膜达45%,在等电点pI4~7、相对分子质量(14.40~116.00)×103范围内,分离得到正常早孕绒毛蛋白点大约680个,蜕膜蛋白点大约621个,其中至少33个蛋白点在绒毛中的表达量高于蜕膜3倍以上,27个蛋白点在蜕膜中的表达量高于绒毛3倍以上,利用质谱相关技术鉴定出7个有意义的差异表达蛋白点。结论早期胚胎绒毛和蜕膜蛋白质的差异表达,可能是胚胎植入的一个原因。
OBJECTIVE: To analyze the differential expression of proteome in villus and deciduas of early embryos in early stage of proteome, and to explore the possible mechanism of embryo implantation. Methods From January to May 2007, 20 cases of embryo villi and uterine decidua in induced abortion were selected in our hospital for 10 weeks. Solid phase pH gradient two-dimensional gel electrophoresis (2D-PAGE) was used to separate the proteome of the embryo villi and decidua in normal early pregnancy induced abortion. Coomassie brilliant blue color, PDQuest2DE software analysis. Results Image analysis showed that the matching rate of chorionic villi and decidua gel was 45%, and in the range of pI4 ~ 7 and relative molecular weight (14.40 ~ 116.00) × 103, A total of 621 decidual proteins were found, of which at least 33 protein spots were more than 3 times higher than that of the decidua and 27 were more than 3 times higher than that of the chorion. The related art identified seven significant differentially expressed protein spots. Conclusion The differential expression of early embryo villi and decidual proteins may be one of the reasons for embryo implantation.
