目的观察以一种主要成分为苄索氯铵和异丙醇的口腔印模专用消毒剂对口腔印模表面乙肝病毒活性及免疫原性的影响。方法使用消毒液对HBV污染的口腔印模进行喷雾消毒,检测HBs Ag含量和HBV-DNA含量,评价消毒效果;检测消毒前后HBV在体外诱导淋巴细胞分泌IFN-γ和IL-2含量,评价对HBV免疫能力的影响。结果喷雾消毒3 min和15 min,对HBs Ag和HBV-DNA的灭活效果明显高于消毒1 min组,灭活率分别为(98.8±1.2)%、(98.9±1.1)%和(99.3±1.1)%、(99.4±1.3)%。消毒3 min和15 min HBV诱导淋巴细胞IFN-γ相对表达量明显高于空白对照和未消毒组;与空白细胞组比较,未消毒组HBV诱导淋巴细胞IL-2相对表达量明显增加,而各消毒组的IL-2相对表达量未发生显著变化。结论该消毒剂喷涂口腔印模时,对HBV的最佳消毒时间为3 min。此时HBV仍然保持较强的免疫原性,能够诱导淋巴细胞IFN-γ表达水平上调,对IL-2表达水平无影响。
Objective To observe the effect of oral sterilizing agent with benzethonium chloride and isopropanol as the main component on the activity and immunogenicity of hepatitis B virus on the surface of dental impressions. Methods The disinfectant solution was used to disinfect the oral impressions of HBV contamination. The contents of HBs Ag and HBV-DNA were detected to evaluate the disinfection effect. The levels of IFN-γ and IL-2 secreted by HBV in vitro were detected before and after disinfection Influence of HBV immunity. Results The inactivation rates of HBsAg and HBV-DNA were significantly higher than that of the disinfection group 1 min after spraying for 3 min and 15 min, respectively (98.8 ± 1.2%, 98.9 ± 1.1% and 99.3 ± 1.1)%, (99.4 ± 1.3)%. Compared with blank control group and non-sterile control group, the relative expression level of IFN-γ in lymphocytes induced by HBV during 3 min and 15 min disinfection was significantly higher than that of blank control group The relative expression of IL-2 in the disinfection group did not change significantly. Conclusion The optimum disinfection time for HBV when the disinfectant is sprayed on the oral impression is 3 min. At this time, HBV still maintained strong immunogenicity, and could induce the up-regulation of IFN-γ expression in lymphocytes, but had no effect on the expression of IL-2.
