目的:研究BID和CTNNB1基因在中药复方理冲生髓饮诱导人卵巢癌细胞株SKOV3细胞凋亡中的作用。方法:Wistar大鼠随机分组,灌胃给药,采血制备中药复方药物血清。提取对照组及理冲生髓饮组肿瘤细胞mRNA,制备DNA探针并分别用Cy3和Cy5两种荧光染料标记,与人细胞凋亡寡核苷酸微阵列芯片进行杂交,经扫描、分析得出药物作用后表达有差异的基因。结果:共筛选出显著表达差异基因66种,其中20种基因表达水平上调,46种基因表达水平下调。结论:理冲生髓饮可能通过上调BID基因和下调CTNNB1基因达到诱导细胞凋亡、抑制转移及增加化疗敏感性的目的。
Objective: To investigate the role of BID and CTNNB1 genes in the apoptosis of human ovarian cancer cell line SKOV3 induced by Li Chongshengyin capsule. Methods: Wistar rats were randomly divided into three groups: gavage and blood sampling to prepare Chinese medicine compound drug serum. The mRNA of tumor cells was extracted from the control group and the Li Chong Sheng Su Yin Beverage Group. DNA probes were prepared and labeled with two fluorescent dyes, Cy3 and Cy5, respectively, and then hybridized with human apoptotic oligonucleotide microarray.
