目的建立同时测定畅鼻通颗粒中桂皮酸、黄芩苷和黄芩素含量的反相高效液相色谱法。方法采用反相高效液相色谱法进行含量测定。色谱柱为DiamonsilTMC18(4.6 mm×200 mm,5μm)色谱柱,流动相为甲醇-乙腈-体积分数为1%的冰醋酸水溶液(体积比为65∶20∶115),流速为1.0 mL.min-1,检测波长为280 nm。结果桂皮酸在0.82~8.20 mg.L-1内,黄芩苷在31.00~310.00 mg.L-1内,黄芩素在3.20~32.00 mg.L-1内,质量浓度与峰面积线性关系良好,相关系数分别为0.999 9、0.999 9和0.999 7;平均回收率分别为101.1%、100.5%和101.4%;精密度RSD均小于2.0%。结论RP-HPLC法可用于畅鼻通颗粒的质量控制。
Objective To establish a method for simultaneous determination of cinnamic acid, baicalin and baicalein in Changbitong Granules by RP-HPLC. Methods The content was determined by reversed-phase high performance liquid chromatography. The column was a DiamonsilTM C18 (4.6 mm×200 mm, 5 μm) column with a mobile phase of methanol-acetonitrile-1% by volume glacial acetic acid (65:20:15 vol/vol) flow rate of 1.0 mL.min- 1, the detection wavelength is 280 nm. Results The concentration of cinnamic acid in the range of 0.82~8.20 mg.L-1, the concentration of baicalin in 31.00~310.00 mg.L-1, and the concentration of baicalein in the range of 3.20~32.00 mg.L-1 showed a good linear relationship between mass concentration and peak area. The coefficients were 0.999 9, 0.999 9, and 0.999 7. The average recoveries were 101.1%, 100.5%, and 101.4%, respectively; the RSDs were all less than 2.0%. Conclusion RP-HPLC can be used to control the quality of Changjingtong Granules.
