目的探讨Chk1表达与食管鳞癌浸润转移的关系。方法采用Western Blot、RT-PCR技术分别检测62例食管鳞癌组织、31例癌旁不典型增生组织及62例正常食管黏膜组织中Chk1蛋白及mRNA的表达,探讨Chk1表达与食管鳞状细胞癌浸润转移之间的关系。结果正常食管黏膜组织、癌旁不典型增生组织以及食管鳞癌组织中,Chk1蛋白、mRNA的表达水平依次升高,三者间两两比较差异均有统计学意义(P<0.05);深层浸润组食管鳞癌组织中Chk1蛋白、mRNA的表达水平均显著高于浅层浸润组,两组比较差异有统计学意义(P<0.05);有淋巴结转移组的食管鳞癌组织中Chk1蛋白、mRNA的表达水平显著高于无淋巴结转移组,两组比较差异有统计学意义(P<0.05)。结论 Chk1高表达与食管鳞癌浸润转移有关。
Objective To investigate the relationship between Chk1 expression and invasion and metastasis of esophageal squamous cell carcinoma. Methods The expressions of Chk1 protein and mRNA in 62 cases of esophageal squamous cell carcinoma, 31 cases of paraneoplastic atypical hyperplasia tissues and 62 cases of normal esophageal mucosa tissues were detected by Western Blot and RT-PCR respectively. Results The expression levels of Chk1 protein and mRNA in normal esophageal mucosa, paraneoplastic atypical hyperplasia and esophageal squamous cell carcinoma were increased in sequence, with significant differences between the two groups (P <0.05). The deep infiltration The expression of Chk1 protein and mRNA in esophageal squamous cell carcinoma was significantly higher than that in superficial invasive group, the difference was statistically significant (P <0.05). Chk1 protein and mRNA in esophageal squamous cell carcinoma with lymph node metastasis (P <0.05). The difference between the two groups was statistically significant (P <0.05). Conclusion Chk1 overexpression is associated with invasion and metastasis of esophageal squamous cell carcinoma.
