建立了化学-海因酶法制备光学纯D-茶氨酸的新方法。采用氰酸盐和L-谷氨酸关环制备D,L-羧乙基海因,与乙胺进行侧链衍生后得到酶转化底物N-乙基海因丙酰胺;进而采用D-海因酶催化水解得到N-氨甲酰-D-茶氨酸,经亚硝酸水解最终得到标题化合物。总收率达55%。对海因酶催化和亚硝酸水解过程条件进行了优化。
A new method of preparing optically pure D-theanine by chemical-hydratase was established. D, L-carboxyethyl hydantoin is prepared by ring closure with cyanate and L-glutamic acid, and the side chain derivatization with ethylamine is used to obtain the enzyme conversion substrate N-ethylhydantoin propionamide; and further the D-sea N-carbamoyl-D-theanine was obtained by enzymatic hydrolysis and was hydrolyzed by nitrous acid to give the title compound. The total yield of 55%. The conditions of hydantoinase and nitrous acid hydrolysis were optimized.
