建立了HPLC-ELSD测定醉鱼草果实中三萜类成分含量的方法。采集不同产地醉鱼草果实药材共9批,采用反相高效液相色谱-蒸发光散射检测法测定其三萜成分。色谱条件:Waters Sun FireTMC18柱(4.6 mm×150 mm,5μm),流动相为甲醇-水(82∶18)溶液,体积流量1 m L·min-1,柱温30℃。ELSD参数:漂移管温度106℃,喷雾温度50℃,载气(N2)流速1.5 L·min-1,增益系数1。结果表明醉鱼草果实中的3种三萜成分clinoposaponinⅢ,desrhamnoverbascosaponin,mimengosideⅠ的进样量分别在0.702~28.08μg(r=0.999 2),0.390~15.60μg(r=0.998 9),0.192~7.68μg(r=0.999 0)呈良好线性关系;平均加样回收率(n=6)分别为99.41%,99.08%,98.67%,相对标准偏差分别为0.86%,1.6%,1.8%。该方法简便、准确、可靠,适用于测定醉鱼草果实中三萜类成分的含量。
A method for the determination of triterpenoids in the fruit of Buddleja by HPLC-ELSD was established. A total of 9 batches of medicinal materials of Buddleja from different areas were collected and their triterpenoids were determined by RP-HPLC. Chromatographic conditions: a Waters Sun FireTMC18 column (4.6 mm × 150 mm, 5 μm) with a mobile phase of methanol-water (82:18) at a volume flow rate of 1 mL min -1 and a column temperature of 30 ° C. ELSD parameters: drift tube temperature 106 ℃, spray temperature 50 ℃, carrier gas (N2) flow rate 1.5 L · min-1, gain factor 1. The results showed that the injection rates of clinoposaponinⅢ, desrhamnoverbascosaponin and mimengosideⅠof three kinds of triterpenes in the fruit of Buddleja were 0.702-28.08μg (r = 0.999 2), 0.390-15.60μg (r = 0.998 9) and 0.192-7.68μg (r = 0.999 0). The average recoveries (n = 6) were 99.41%, 99.08% and 98.67%, respectively. The relative standard deviations were 0.86%, 1.6% and 1.8% respectively. The method is simple, accurate and reliable and is suitable for the determination of triterpenoids in the fruit of Buddleia.
