新型复合式肝损伤大鼠模型的建立

目的建立一种由猪血清、四氯化碳、乙醇3种因素刺激的新型肝损伤大鼠模型。方法将56只健康雄性大鼠随机分为A、B、C、D、E 5组,其中A至D组为实验组,E为空白对照组。除空白对照组外,其余各组连续4周多因素诱导肝损伤模型。每周摘眼球取血,测定血清中谷草转氨酶(AST)、谷丙转氨酶(ALT)活性。第4周取血后处死动物,取肝脏进行切片观察。结果多因素诱导肝损伤各模型组AST和ALT均不同程度的升高,与空白对照组相比,在不同时间点上有显著性差异(P<0.05)或非常显著性差异(P<0.01)。病理切片也证实各实验组出现不同程度的肝损伤。结论由猪血清、四氯化碳、乙醇3种因素诱导可建立较为理想的大鼠肝损伤模型。 Objective To establish a rat model of liver injury induced by three factors of pig serum, carbon tetrachloride and ethanol. Methods 56 healthy male rats were randomly divided into A, B, C, D and E 5 groups, of which A to D were experimental groups and E was blank control group. Except for the blank control group, the other groups were induced by multiple factors for 4 weeks. Blood was taken from the eyeball every week to determine serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activity. Animals were sacrificed on the fourth week after the blood was taken, and the liver was sectioned for observation. Results Compared with the blank control group, the levels of AST and ALT increased significantly in various models of liver injury induced by multiple factors (P <0.05) or very significant difference (P <0.01) . Pathological section also confirmed that the experimental group showed varying degrees of liver injury. Conclusion The rat model of liver injury induced by three factors of pig serum, carbon tetrachloride and ethanol can be established.
