目的:建立离子对反相高效液相色谱法测定血浆中百草枯的浓度。方法:采用Discovery C_(18)色谱柱;流动相为乙腈-庚烷磺酸钠溶液(12∶88);10%三氯醋酸直接沉淀血浆蛋白;流速为1.0 m L·min~(-1);检测波长为258 nm。结果:定量下限为0.1μg·m L~(-1),线性范围为0.1~10.0μg·m L~(-1),批内和批间RSD分别为3.77%和10.60%,平均回收率大于89.31%。结论:本法具有灵敏、准确、稳定、操作简便等优点,符合生物样本测定的要求,适用于测定人血浆中百草枯的浓度。
Objective: To establish an ion-pair reversed-phase high performance liquid chromatographic method for the determination of paraquat in plasma. Methods: The mobile phase consisted of acetonitrile - heptane sulfonate solution (12:88), and the plasma protein was directly precipitated with 10% trichloroacetic acid at a flow rate of 1.0 m L · min -1 using Discovery C 18 column. The detection wavelength is 258 nm. Results: The lower limit of quantification was 0.1 μg · m L -1 and the linear range was 0.1-10.0 μg · m L -1. The intra-and inter-assay RSD were 3.77% and 10.60% respectively. The average recovery was greater than 89.31%. Conclusion: This method is sensitive, accurate, stable, easy to operate and other advantages, in line with the requirements of biological samples for determination of paraquat in human plasma concentration.
