高效薄层色谱法(HPTLC)是近年来发展起来的新的薄层技术。本文介绍了HPTLC的理论基础、实验技术及其应用。理论方面讨论了理论塔板数与R_f的关系及分离度。对实验技术包括高效再层板、点样、展开方法、斑点的显色及薄层色谱扫描定量法等作了较为详细的探讨。应用方面介绍了巴比妥类药物、镇痛退热药及前列腺素制剂等药物及其制剂的定量分析,药物中微量杂质的控制以及体液中药物浓度及代谢产物的分离测定等。
High-performance thin-layer chromatography (HPTLC) is a new thin-layer technology developed in recent years. This article introduces the theoretical basis, experimental techniques and applications of HPTLC. Theoretical aspects discuss the relationship between the number of theoretical plates and R_f and the degree of separation. The experimental techniques, including high-efficiency re-laminating, spotting, unfolding methods, spot coloration, and thin-layer chromatography scanning quantitative methods, were discussed in detail. Application aspects include the quantitative analysis of drugs and preparations including barbiturates, analgesic antipyretics and prostaglandin preparations, control of trace impurities in drugs, and determination of drug concentrations and metabolites in body fluids.
