目的探讨银杏叶提取物(EGb)对糖尿病大鼠主动脉糖基化终产物受体mRNA表达的调节作用。方法Wistar大鼠喂以高糖高脂饲料加链脲佐菌素(Streptozotocin,STZ)建立的2型糖尿病大鼠模型。将Wistar大鼠随机分为正常对照组、2型糖尿病模型组、银杏叶提取物治疗组及氨基胍治疗组。12周末应用原位杂交法观察大鼠主动脉内皮细胞糖基化终产物受体mRNA表达。结果2型糖尿病模型组大鼠主动脉糖基化终产物受体mRNA表达增加,明显高于对照组(P<0.01);银杏叶提取物治疗组及氨基胍治疗组大鼠主动脉糖基化终产物受体mRNA表达降低,低于模型组(P<0.05)。结论银杏叶提取物能抑制糖尿病大鼠主动脉糖基化终产物受体mRNA表达,对高血糖及糖基化终产物(AGE)造成主动脉血管损伤具有保护作用。
Objective To investigate the effect of ginkgo biloba extract (EGb) on the mRNA expression of aglycosylated end products in diabetic rats. Methods Wistar rats were fed a high-sugar high-fat diet plus streptozotocin (STZ) to establish a rat model of type 2 diabetes. Wistar rats were randomly divided into normal control group, type 2 diabetes model group, Ginkgo biloba extract treatment group and aminoguanidine treatment group. At the end of 12 weeks, in situ hybridization was used to observe the expression of receptor mRNA of glycosylation end products in rat aortic endothelial cells. Results The mRNA expression of aglycin endproducts in type 2 diabetes mellitus rats was increased, which was significantly higher than that in the control group (P<0.01). Glycosylation of aorta in Ginkgo biloba extract group and aminoguanidine treatment group rats The expression of end-product receptor mRNA was decreased and was lower than that of the model group (P<0.05). Conclusion Ginkgo biloba extract can inhibit the mRNA expression of aglycosylated end products in diabetic rats, and it has a protective effect on aortic vascular injury caused by hyperglycemia and advanced glycation end products (AGE).
