目的 对脑血管内皮细胞中神经肽 Y(NPY)的 Y1 受体进行精确定位 .方法 用针对 NPY的 Y1 受体 C末端肽片段的一种纯化的抗血清 ,用免疫组织化学方法和包埋前免疫金 -银加强法观察 Y1 受体在脑血管内皮细胞中的分布 .结果 大脑皮质和髓质、丘脑、下丘脑、小脑和脑干的微血管有散在的含 Y1 受体免疫反应的 (- Ir)内皮细胞 ,孤立的脑底动脉及其分支的一些内皮细胞也具有 Y1 受体免疫反应性 (- L I) ,Y1 受体 - L I分布于内皮细胞的血管腔面和平滑肌细胞面的细胞膜和胞质中 .结论 来源于血液、内皮细胞和血管周围神经的 NPY通过内皮细胞上 Y1 受体的介导 ,可能参与对脑血流的调节和促进血管增生
Objective To accurately locate the Y1 receptor of neuropeptide Y (NPY) in cerebrovascular endothelial cells.METHODS A purified antiserum against the Y1 receptor C-terminal peptide fragment of NPY was used for immunohistochemical staining Immuno gold-silver staining was used to observe the distribution of Y1 receptor in cerebrovascular endothelial cells.Results Microvessels in the cerebral cortex and medulla, the thalamus, hypothalamus, cerebellum and brain stem were scattered with Y1 receptor-reactive (- Ir ) Endothelial cells, isolated basilar artery and some endothelial cells of their branches also have Y1 receptor immunoreactivity (- LI), Y1 receptor - LI distributes in the luminal and smooth muscle cell surface of endothelial cells Quality.Conclusion The NPY derived from blood, endothelial cells and perivascular nerves is mediated by Y1 receptors on endothelial cells and may be involved in the regulation of cerebral blood flow and the promotion of vascular proliferation
