目的:观察鳖甲煎丸对免疫性肝纤维化模型大鼠肝组织肝细胞生长因子(HGF)表达的影响,分析鳖甲煎丸抗肝纤维化的治疗作用并探讨其作用机制。方法:雄性Wistar大鼠90只,随机分为6组:正常对照组、模型组、秋水仙碱预防组、秋水仙碱治疗组、鳖甲煎预防组、鳖甲煎治疗组,采用猪血清诱导免疫损伤性肝纤维化模型。各组于10周后随机取8只大鼠断头处死,取血清,检测血清透明质酸(HA)、层黏连蛋白(LN)、Ⅲ型前胶原(PCⅢ)、Ⅳ型胶原(Ⅳ-C);取肝组织,免疫组化染色,用图像分析法检测HGF。结果:各预防组、治疗组血清HA,LN,PCⅢ,Ⅳ-C含量均明显降低(P<0.01或P<0.05),但以鳖甲煎预防组、治疗组降低最为明显;各预防组、治疗组大鼠肝脏组织HGF均有表达(P<0.01或P<0.05),鳖甲煎预防组、治疗组比秋水仙碱组的表达更为明显。结论:鳖甲煎丸能够抑制肝脏纤维化病理改变,增加HGF的表达,具有明显的抗肝纤维化作用。
Objective: To observe the effect of Biejiajian Pills on the expression of hepatocyte growth factor (HGF) in liver tissue of rats with autoimmune hepatic fibrosis and to analyze its therapeutic effect on hepatic fibrosis and its mechanism. Methods: Ninety male Wistar rats were randomly divided into 6 groups: normal control group, model group, colchicine prevention group, colchicine treatment group, Biejiajian prevention group and Biejiajian treatment group, which were induced by porcine serum Immune-damaging liver fibrosis model. After 10 weeks, 8 rats in each group were decapitated and sacrificed. Serum samples were taken for determination of serum HA, laminin (LN), PCⅢ, Ⅳ- C); liver tissue was taken for immunohistochemical staining, and HGF was detected by image analysis. Results: The levels of HA, LN, PCⅢ, Ⅳ-C in all preventive and treatment groups were significantly decreased (P <0.01 or P <0.05), but the prevention group was the best, The expression of HGF in the liver of the treated group was significantly higher than that of the BCC group (P <0.01 or P <0.05). Conclusion: BJJ can inhibit the pathological changes of hepatic fibrosis, increase the expression of HGF, and has a significant anti-hepatic fibrosis effect.
