目的 :确定P - 170糖蛋白在多药耐药 (MDR)K5 6 2 /ADM细胞中表达的位置。方法 :通过免疫反应并利用流式细胞仪 (FCM )检测表达P - 170糖蛋白细胞的百分含量 ;用免疫电镜技术检测P- 170糖蛋白在细胞中的位置及其表达水平。结果 :敏感株K5 6 2细胞与耐药株K5 6 2 /ADM细胞对P - 170糖蛋白的表达有非常明显的差异 ;不同的K5 6 2 /ADM细胞表达P - 170糖蛋白的量差别较大 ;少量的K5 6 2细胞也有极少的P - 170糖蛋白的表达 ;细胞表达的P - 170糖蛋白位于细胞膜上。结论 :多药耐药K5 6 2 /ADM细胞表达的P - 170糖蛋白最终被镶嵌在细胞膜上 ;在该蛋白质从被表达、修饰和转运到细胞膜上的过程中 ,该蛋白质不具有成熟P - 170糖蛋白的特异构象
Purpose: To determine the location of P-170 glycoprotein in multidrug resistance (MDR) K562/ADM cells. Methods: The percentage of P-170 glycoprotein expressed in cells was detected by flow cytometry (FCM); the position and expression level of P-170 glycoprotein in cells were detected by immunoelectron microscopy. Results: There was a significant difference in the expression of P-170 glycoprotein between K562 cells and K562/ADM cells. The difference of P-170 glycoprotein expression between K562/ADM cells was different. Large; a small amount of K562 cells also have very little P-170 glycoprotein expression; cells express the P-170 glycoprotein located on the cell membrane. CONCLUSIONS: The P-170 glycoprotein expressed in multidrug resistant K562/ADM cells is eventually mounted on the cell membrane; the protein does not have mature P- during the process of expression, modification and transport from the protein to the cell membrane.
