目的:以输卵管妊娠的输卵管标本为研究对象,观察钙结合蛋白Calbindin-d28k(CaBP-d28k)在输卵管妊娠输卵管上皮组织中的表达变化,探讨CaBP-d28k与输卵管妊娠的关系。方法:30例既往月经规律、妊娠囊尚未破裂的输卵管妊娠患者,每例患者取材均包括输卵管妊娠受精卵种植部位和非种植部位的输卵管标本。经HE染色进行组织学观察确定种植部位,采用免疫组织化学法和逆转录聚合酶链反应法检测CaBP-d28k在输卵管妊娠种植部位和非种植部位输卵管上皮中的蛋白和基因表达。结果:30例输卵管妊娠非种植部位的输卵管黏膜上皮细胞均可见CaBP-d28k蛋白及CaBP-d28kmRNA强阳性表达。种植部位的输卵管黏膜上皮细胞CaBP-d28k蛋白及CaBP-d28kmRNA表达较非种植部位明显减少(P<0.05),有些种植部位CaBP-d28k蛋白表达甚至缺如。结论:输卵管妊娠种植部位的输卵管黏膜上皮细胞CaBP-d28k蛋白及CaBP-d28kmRNA表达较非种植部位明显减少,推测CaBP-d28k与输卵管妊娠的发生可能存在密切联系。
OBJECTIVE: To investigate the expression changes of Calbindin-d28k (CaBP-d28k) in tubal pregnancy tubal epithelial tissues and to investigate the relationship between CaBP-d28k and tubal pregnancy. Methods: Thirty patients with tubal pregnancy who had no previous history of menstrual cycle and no ruptured gestational sac were enrolled in this study. Tubal specimens were collected from the implantation site and non-implantation site of the fertilized oviduct in each patient. Histopathological examination of HE staining to determine the site of implantation, immunohistochemistry and RT-PCR were used to detect the protein and gene expression of CaBP-d28k in the tubal epithelium of the implantation site and non-implantation site of tubal pregnancy. Results: Strong expression of CaBP-d28k protein and CaBP-d28kmRNA were observed in the oviduct epithelial cells of 30 cases of tubal pregnancy. The expression of CaBP-d28k protein and CaBP-d28kmRNA in tubal mucosal epithelial cells at implantation site was significantly lower than that at non-implantation site (P <0.05), and even at some sites. CONCLUSIONS: The expression of CaBP-d28k protein and CaBP-d28kmRNA in tubal epithelial cells at the implantation site of tubal pregnancy is significantly lower than that at non-implantation site. It is speculated that there may be a close relationship between CaBP-d28k and tubal pregnancy.
