[目的]观察大株红景天注射液抗心肌缺氧/复氧损伤的保护作用,并探讨其抗心肌缺氧/复氧可能的机制。[方法]原代培养的心肌细胞随机分为5组:空白对照组;缺氧/复氧组;缺氧/复氧分别加大株红景天注射液5、10、20 m L/L组。加药孵育12 h后,缺氧培养9 h,随后常氧培养2 h。噻唑蓝(MTT)法检测细胞增殖,CMH2DCF-DA荧光探针检测细胞内活性氧簇(ROS)的生成量,JC-1荧光探针检测细胞内线粒体膜电位,以及测定细胞内超氧化物歧化酶SOD活力及其基因表达量。[结果]大株红景天注射液增加了缺氧/复氧损伤心肌细胞的活力,减少细胞内ROS的生成,抑制细胞内线粒体膜电位的降低,同时增加了心肌细胞内SOD的活力及其基因表达量。[结论]大株红景天注射液对缺氧/复氧(H/R)损伤的心肌细胞有保护作用。
[Objective] To observe the protective effect of Rhodiola rosea against myocardial hypoxia / reoxygenation injury and to explore its possible mechanism of anti-myocardial hypoxia / reoxygenation. [Methods] Primary cultured cardiomyocytes were randomly divided into 5 groups: blank control group, hypoxia / reoxygenation group, hypoxia / reoxygenation group were given Rhodiola injection 5,10,20 m L / L group . After dosing for 12 h, the cells were cultured in hypoxia for 9 h and then cultured in normoxia for 2 h. The cell proliferation was detected by MTT assay. The production of reactive oxygen species (ROS) in cells was detected by CMH2 DCF-DA fluorescent probe. The mitochondrial membrane potential was detected by JC-1 fluorescent probe and the intracellular superoxide dismutase SOD activity and its gene expression. [Result] Rhizoma Rhodiola injection increased the activity of hypoxia / reoxygenation injury cardiomyocytes, decreased intracellular ROS production, inhibited the reduction of mitochondrial membrane potential and increased the activity of SOD in cardiomyocytes Gene expression level. [Conclusion] Rhizoma Rhodiolae has a protective effect on cardiomyocytes injured by hypoxia / reoxygenation (H / R).
