本文应用细胞因子依赖细胞株CTLL和7TD1及。~3H-TDR掺入方法,分别对6种常用培养杂交瘤的饲养细胞培养上清和条件培养液中IL-2和IL-6的生物学活性水平进行了定量检测和比较。结果发现,除大鼠混合胸腺细胞培养上清中含有约1u/ml IL-2外,其它均未测出IL-2的生物学活性。但6种上清中都含有IL-6,其中人成纤维细胞CRL_(1506)和人内皮细胞中含有很高水平的IL-6,分别为10000u/ml和2512u/ml;大鼠混合胸腺细胞和小鼠腹腔巨噬细胞培养上清分别为158u/ml和126u/ml;小鼠脾脏细胞和胸腺细胞培养上清中IL-6相对较低,分别为79u/ml和16u/ml。此外,本文还对常用饲养细胞及条件培养液在杂交瘤细胞生长中可能起的主要作用进行了讨论。
This article applies cytokine-dependent cell lines CTLL and 7TD1 and. With the ~3H-TDR incorporation method, the biological activity levels of IL-2 and IL-6 in the culture supernatant and the conditioned medium of six commonly used hybridoma cultured feeders were quantitatively examined and compared. As a result, it was found that the biological activity of IL-2 was not measured except that the culture supernatant of rat mixed thymocytes contained approximately 1 u/ml IL-2. However, the six supernatants contained IL-6, among which human fibroblasts CRL_(1506) and human endothelial cells contained very high levels of IL-6, which were 10,000 u/ml and 2512 u/ml, respectively; rat mixed thymocytes The culture supernatants of mouse and mouse peritoneal macrophages were 158u/ml and 126u/ml, respectively; the IL-6 levels in mouse spleen cells and culture supernatants of thymus cells were relatively low, being 79u/ml and 16u/ml, respectively.
