目的:观察新型溶瘤腺病毒M1在体内代谢的时间及对肿瘤生长的抑制作用。方法:建立人胃癌裸鼠原位移植瘤动物模型,在指定时间点处死动物,观察瘤体大小,分离原发瘤及转移瘤进行病毒滴度测定,采用免疫组化及原位杂交来检测病毒的分布和活性复制。结果:在人胃癌裸鼠原位模型上,M1经静脉输注后不仅分布于胃原发瘤,而且存在于转移灶,但用M1静脉内给药可以减缓肿瘤生长速度。结论:静脉内输注溶瘤腺病毒可以选择性的作用于肿瘤及转移灶,并发挥溶瘤和靶向灭活plk1的双重效应。
OBJECTIVE: To observe the time of metabolism of new oncolytic adenovirus M1 in vivo and its inhibitory effect on tumor growth. Methods: The animal model of orthotopic transplantation of human gastric cancer in nude mice was established. The animals were sacrificed at the indicated time points. The tumor size was observed. The primary tumors and metastases were isolated for virus titer determination. Immunohistochemistry and in situ hybridization were used to detect the virus The distribution and activity of replication. Results: In the in situ model of gastric cancer in nude mice, M1 was not only distributed in the primary gastric tumor but also in the metastatic tumor after intravenous infusion. However, the intramuscular administration of M1 could reduce the tumor growth rate. CONCLUSION: Intravenous infusion of oncolytic adenovirus can selectively act on tumors and metastases and exert the dual effects of oncolytic and targeted inactivation of plk1.
