目的:观察油酸(oleic acid,OA)肺损伤对大鼠肝组织白细胞介素-1β(IL-1β)、白细胞介素-10(IL-10)表达的影响。方法:16只健康Wistar大鼠随机分为对照组、OA组,每组各8只大鼠。采用OA法复制急性呼吸窘迫综合征(ARDS)模型。观察2组大鼠肺湿干重比值(W/D)及肝功能指标丙氨酸氨基转移酶(ALT)、天门冬酸氨基转移酶(AST)、胆红素(TB)的改变,光镜下观察肝组织病理形态学改变,检测肝组织中IL-1β、IL-10蛋白及mRNA表达。结果:OA组W/D、ALT、AST、TB均高于对照组(P<0.01);OA组肝组织病理形态学结构较对照组肝细胞弥漫性水肿,肝小叶中央静脉周围肝细胞胞浆疏松化,小叶周围肝细胞胞浆空泡化,小叶内部分肝细胞核消失,肝小叶中央静脉和肝血窦充血等改变;OA组肝组织中IL-1β、IL-10蛋白及IL-1β、IL-10mRNA含量的表达均较对照组增强(P<0.05)。结论:OA性ARDS并发肝损伤的机制中,可能有IL-1β、IL-10的参与。
Objective: To observe the effect of oleic acid (OA) on the expression of interleukin-1β (IL-1β) and interleukin-10 (IL-10) in rat liver tissue. Methods: Sixteen healthy Wistar rats were randomly divided into control group and OA group with 8 rats in each group. Acute respiratory distress syndrome (ARDS) model was replicated by using OA method. The wet / dry weight ratio (W / D) and the changes of liver function indexes such as alanine aminotransferase (ALT), aspartate aminotransferase (AST) and bilirubin (TB) Under the pathological changes of liver tissue, the expression of IL-1β, IL-10 protein and mRNA in liver tissue were detected. Results: The W / D, ALT, AST and TB in OA group were higher than those in control group (P <0.01). The histopathological features of liver tissue in OA group were more diffuse edema than those in control group, Loosening, cytoplasmic vacuolization of hepatocytes around the leaflets, disappearance of part of the hepatocytes in the lobules, congestion of the hepatic lobule central veins and hepatic sinusoids. The levels of IL-1β, IL-10 and IL-1β, The expression of IL-10mRNA increased compared with the control group (P <0.05). Conclusion: The mechanism of OA-induced ARDS complicated by liver injury may involve IL-1β and IL-10.
