应用酵母双杂交技术, 鉴定Wnt通路关键信号分子TCF4与cAMP效应元件结合蛋白家族新成员—— 激活转录因子5(ATF5)之间的相互作用, 酵母表型分析和b-半乳糖苷酶(b-gal)活性测定结果发现, 两者的作用位于ATF5蛋白C端含有碱性亮氨酸拉链(bZIP)区域(162 ~ 282 aa). 通过受TCF启动的荧光酶报告系统分析ATF5对Wnt信号通路的影响, 结果显示ATF5蛋白C端能进一步加强TCF4对Wnt通路的激活效应. 由此提示, ATF5可能是Wnt通路下游信号的辅助激活因子(co-activator), 它能协同 b-连环蛋白/TCF4参与Wnt信号通路的激活.
The yeast two-hybrid assay was used to identify the interaction between TCF4, a key signaling molecule of Wnt signaling pathway, and ATF5, a novel member of the cAMP response element binding protein family, yeast phenotype analysis and b-galactosidase (b -gal activity assay showed that both of them play a role in the bZIP region (162-282 aa) at the C-terminus of ATF5 protein.According to the luciferase reporter system activated by TCF, the effect of ATF5 on Wnt signaling pathway The results showed that ATF5 C-terminal can further enhance the activation of Wnt pathway by TCF4, suggesting that ATF5 may be a co-activator of downstream signaling of Wnt pathway, which can cooperate with b-catenin / TCF4 Involves activation of Wnt signaling pathway.
