目的:研究跑台运动对废用性骨质疏松的作用以及破骨细胞分化OPG-RANKL-RANK系统在其中的介导作用。方法:6周龄雄性Sprague-Dawley大鼠40只,分为正常对照组、废用模型组、正常恢复组和运动恢复组,每组10只。正常对照组不做任何特殊处理,安静饲养4周后处死;废用模型组尾部悬吊4周后处死;正常恢复组尾部悬吊4周后,安静饲养4周处死;运动恢复组尾部悬吊4周后,跑台训练4周处死。各组大鼠处死后立即进行骨密度(BMD)、骨组织形态计量学、骨组织抗酒石酸酸性磷酸酶5b(TRACP-5b)染色计量、骨代谢以及破骨细胞分化OPG-RANKL-RANK系统相关细胞因子等指标的测试。结果:废用模型组大鼠BMD、骨小梁体积百分比(TBV)、骨小梁宽度(Tb.Wi)、血清碱性磷酸酶(ALP)浓度、骨髓细胞因子OPG基因表达量均显著小于正常对照组(P<0.01),而骨小梁间距(Tb.Sp)、胫骨干骺端TRACP-5b平均阳性染色面积百分比(APSAP)、血清Ca2+和TRACP-5b浓度、骨髓细胞因子RANKL、M-CSF、RANK、IL-6和TNF-α基因表达量则显著大于正常对照组(P<0.01)。运动恢复组大鼠BMD、TBV、Tb.Wi、血清ALP浓度、骨髓细胞因子OPG基因表达量均显著大于正常恢复组(P<0.05或P<0.01),而Tb.Sp、血清Ca2+和TRACP-5b浓度、胫骨干骺端APSAP、骨髓细胞因子RANKL、M-CSF、RANK、IL-6和TNF-α基因表达量均显著小于正常恢复组(P<0.05,P<0.01)。结论:4周的尾部悬吊可导致大鼠废用性骨质疏松症的发生,跑台运动可以促进废用性骨质疏松症的恢复,这均与骨髓微环境破骨细胞分化OPG-RANKL-RANK系统相关细胞因子的表达有关。
OBJECTIVE: To study the effect of treadmill exercise on disuse osteoporosis and the mediation of osteoclast differentiation OPG-RANKL-RANK system. Methods: Forty male Sprague-Dawley rats aged 6 weeks were divided into normal control group, untreated group, normal recovery group and exercise recovery group, 10 rats in each group. The rats in the normal control group were sacrificed after 4 weeks and were sacrificed after 4 weeks of recuperation. Rats in the model group were sacrificed after 4 weeks’ After 4 weeks, treadmill training was executed 4 weeks. BMD, histomorphometry, TRACP-5b staining, bone metabolism, osteoclast differentiation and OPG-RANKL-RANK system in all groups were measured immediately after sacrifice Cytokines and other indicators of the test. Results: BMD, trabecular volume (TBV), trabecular bone width (Tb.Wi), serum ALP and bone marrow cytokine OPG gene expression in the untreated group were all significantly lower than those in normal group (P <0.01), while the ratio of trabecular bone distance (Tb.Sp), the percentage of positive staining area of TRACP-5b in the tibial metaphysis (APSAP), serum Ca2 + and TRACP-5b concentration, bone marrow cytokines RANKL, M- CSF, RANK, IL-6 and TNF-α gene expression was significantly greater than the normal control group (P <0.01). The levels of BMD, TBV, Tb.Wi, serum ALP and bone marrow cytokine OPG in exercise recovery group were significantly higher than those in normal recovery group (P <0.05 or P <0.01), while Tb.Sp, serum Ca2 + and TRACP- (P <0.05, P <0.01). The mRNA expression levels of APSAP, bone marrow cytokines RANKL, M-CSF, RANK, IL-6 and TNF-α in the tibia metaphysis were significantly lower than those in the normal control group. Conclusion: Tail suspension for 4 weeks can lead to the occurrence of disuse osteoporosis in rats. Treadmill exercise can promote the recovery of disuse osteoporosis, which is related to osteoclast differentiation OPG-RANKL -RANK system-related cytokine expression.
