化学致癌物与致变物经机体代谢活化,可以原形、中间产物、结合物等形式随尿排出。排出物中有的具有活性,有的需在体外通过酶催化激活或裂解重新释放致变体才产生活性。进行尿诱变性检测,能反映机体接触潜在致癌物与致变物的情况和程度。根据尿致变性及其成份的分析,可为化学物质的致癌性判断提供重要的线索。文献报道目前已发现几种致癌物,体外活化Ames测试为阴性,而尿的测试为阳性。以上情况表明尿中致变物分离与检测的重要性。本文应用XAD-2树脂,对尿中已知致变物进行浓缩分离,结合鼠伤寒沙门氏菌回变试验,对尿中致变物检测方法进行了研究,兹将方法与结果介绍於下。
Chemical carcinogens and mutagens metabolic activation by the body, can prototype, intermediate products, conjugates and other forms with the urine. Some of the effluent activity, and some need in vitro by enzymatic activation or lysis re-release of mutants to produce activity. Urine mutagenicity test, can reflect the body contact potential carcinogens and mutagens situation and extent. According to the analysis of urinary degeneration and its components, it can provide important clues for the judgment of carcinogenicity of chemical substances. Reported in the literature has found several carcinogens, in vitro activation Ames test was negative, and urine test was positive. The above shows the importance of separation and detection of urinary mutagens. In this paper, XAD-2 resin was used to enrich and separate the known mutagens in urine. Combined with the back-change test of Salmonella typhimurium, the detection methods of urinary mutagens were studied. The methods and results are introduced below.
