目的:建立红丝线药材的高效液相色谱(HPLC)指纹图谱。方法:采用HPLC法。色谱柱为ODS-C_(18),流动相为甲醇-水(梯度洗脱),流速为1.0 ml/min,检测波长为370 nm,柱温为30℃,进样量为20μl。以2号色谱峰为参照峰,测定11批样品的HPLC图谱,采用《中药色谱指纹图谱相似度评价系统》(2004 A版)进行共有峰指认和相似度评价。结果:11批药材样品HPLC图谱有10个共有峰,相似度均≥0.965。经验证,11批样品HPLC图谱与对照图谱具有较好的一致性。结论:该试验所建立的HPLC指纹图谱专属性强、结果稳定,可作为红丝线药材质量评价的主要依据之一。
Objective: To establish a high performance liquid chromatography (HPLC) fingerprint of red silk medicine. Methods: HPLC method was used. The chromatographic column was ODS-C_ (18). The mobile phase consisted of methanol-water (gradient elution), the flow rate was 1.0 ml / min, the detection wavelength was 370 nm, the column temperature was 30 ℃ and the injection volume was 20μl. The HPLC chromatograms of 11 batches of samples were determined with reference to Peak 2, and the common peak identification and similarity evaluation were carried out using the “Chinese Medicines Chromatographic Fingerprint Similarity Evaluation System” (2004 A). Results: The HPLC profiles of 11 batches of medicinal samples had 10 common peaks with the similarity of ≥0.965. It has been verified that 11 batches of samples have good consistency with the control spectrum. Conclusion: The HPLC fingerprinting established by this experiment is highly specific and stable, which can be used as one of the main evidences for quality evaluation of red silk medicinal materials.
