我们曾以十二烷基硫酸钠—聚丙烯酰胺凝胶电泳(SDS-PAGE),发现烧伤血清中至少有4个异常区带.我们对这些异常区带作了进一步分析,方法是用蛋白免疫印迹法,以12种抗人不同血清蛋白的抗体检测 SDS-PAGE 中的各区带,发现用抗触珠蛋白抗血清所着染的区带与 SDS-PAGE 中的异常区带非常符合。如果将烧伤血清先用离子交换层析分离,再检测所收集的各蛋白峰洗脱液对正常小鼠脾淋巴细胞是否具免疫抑制活性,则发现具免疫抑制活性的洗脱液均出现在第6蛋白峰.若将此具免疫抑制活性的洗脱液,再进行 SDS-PAGE 和免疫印迹分析,则用触珠蛋白抗血清检测所得区带,仍与烧伤血清中发现的异常区带相符.本实验结果显示在烧伤血清中可能有一类似触珠蛋白的物质具有免疫抑制活性.
We used sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) to find at least 4 abnormal bands in the sera of burn patients. We further analyzed these abnormal bands by immunization with protein Western Blotting. The bands of SDS-PAGE were detected by using 12 kinds of anti-human serum antibodies. The bands stained with anti-haptoglobin antiserum were found to be in good agreement with the abnormal bands in SDS-PAGE. If the sera were separated by ion exchange chromatography, and then detected the collection of each protein peak eluate on normal mouse splenic lymphocytes whether immunosuppressive activity, we found immunosuppressive activity of the elution occurred in the first 6 protein peak.If this immunosuppressive activity of the eluate, and then by SDS-PAGE and Western blot analysis, the detection zone with haptoglobin antiserum, still with the burn zone found in the abnormal zone. The results of this experiment show that there may be a haptoglobin-like substance in burn serum that has immunosuppressive activity.
