目的:研究红景天加维甲酸对大鼠胃粘膜癌变DNA含量、C-MYC mRNA表达及胃粘膜细胞凋亡的影响。方法:采用MNNG、酒精及10% NaCl混合液建立大鼠胃癌模型,设立正常对照组、模型组、维甲酸治疗组和红景天加维甲酸治疗组,分别采用Feulgen染色、免疫组化、Western blotting及TUNEL法检测大鼠胃粘膜DNA含量、C-myc基因表达及细胞凋亡。结果:红景天加维甲酸可抑制大鼠胃粘膜的恶性增殖。红景天加维甲酸治疗组C-myc表达减少,与维甲酸治疗组比较有显著性差异(P<0.01)。结论:红景天加维甲酸能下调C-myc基因表达,促进细胞凋亡,这可能是其抗癌的重要机制之一。
Objective: To study the effect of Rhodiola plus Retinoic Acid on DNA content, C-MYC mRNA expression and gastric mucosal cell apoptosis in gastric mucosa of rats. METHODS: A rat gastric cancer model was established with a mixture of MNNG, alcohol and 10% NaCl. The normal control group, the model group, the retinoic acid treatment group, and the Rhodiola plus formic acid treatment group were established. Feulgen staining, immunohistochemistry, and Western were used, respectively. The gastric mucosal DNA content, C-myc gene expression and apoptosis were detected by blotting and TUNEL. RESULTS: Rhodiola plus retinoic acid inhibited the malignant proliferation of gastric mucosa in rats. Rhodiola plus retinoic acid treatment group decreased expression of C-myc, compared with retinoic acid treatment group was significantly different (P <0.01). Conclusion: Rhodiola plus Retinoic acid can down-regulate the expression of C-myc gene and promote apoptosis. This may be one of the important mechanisms of its anti-cancer effect.
