目的 探讨甲状腺转录因子 (TTF 1)在甲状腺肿瘤及滤泡上皮细胞不同部位的表达及与RET、Galectin 3(Gal 3)和粘蛋白 1(MUC1)基因产物的相关性。方法 应用免疫组织化学 (SP)法 ,对 133例甲状腺手术切除标本进行了TTF 1、RET、Gal 3和MUC1的联合检测。结果 (1)TTF 1:在病变 (瘤 )细胞核、质及周围正常组织的细胞核均有表达 ,4种不同病理类型的周围正常滤泡 ,良、恶性病变的胞核阳性表达差别无显著意义 (P >0 0 5 )。而胞质型TTF 1在良性病变的表达率为 2 7 4 % ,恶性肿瘤的表达率为 6 6 1% ,良性病变与两种甲状腺癌组织的差异有显著意义 (P <0 0 5 ) ,恶性肿瘤高于良性病变。(2 )RET :腺瘤组织阳性表达率为 38 9% ,乳头状癌、滤泡性癌阳性表达率分别为 88 4 %和87 5 % ,腺瘤与乳头状癌、滤泡性癌的差异有显著意义 (P <0 0 5 )。 (3)Gal 3、MUC1:良性病变阳性率分别为 38 4 %和 2 3 3% ,恶性肿瘤的阳性率分别为 88 1%和 5 0 9% ,良性病变 (结节性甲状腺肿和腺瘤 )阳性率低于甲状腺乳头状癌和滤泡性癌 (P <0 0 5 ) ;甲状腺癌中Gal 3和MUC1表达基本平行 ,但Gal 3的敏感性高于MUC1;部分腺瘤中可见灶性Gal 3和MUC1同时阳性。而胞质型TTF 1表达与Gal 3、MUC1阳性表达有关。结论 TTF 1胞核反应是正常的
Objective To investigate the expression of thyroid transcription factor (TTF 1) in different parts of thyroid tumors and follicular epithelial cells and their correlation with RET, Galectin 3 (Gal 3) and mucin 1 (MUC1) gene products. Methods Thirty-seven thyroidectomized specimens were examined by immunohistochemistry (SP). The combined detection of TTF 1, RET, Gal 3 and MUC 1 was performed. Results (1) The expression of TTF 1 in the nuclei of the lesions (tumor) nuclei, normal tissues and surrounding normal tissues was also observed. There was no significant difference in the positive expression of the four kinds of normal follicles and the benign and malignant lesions among 4 different pathological types P> 0 0 5). The expression of cytoplasmic TTF 1 in benign lesions was 27.4% and that in malignant tumors was 6 6 1%. There was significant difference between benign lesions and two kinds of thyroid carcinoma (P <0.05) Malignant tumors than benign lesions. (2) RET: The positive rate of adenoma was 38.9%, the positive rate of papillary carcinoma and follicular carcinoma was 88.4% and 87.5%, respectively. The difference between adenoma, papillary carcinoma and follicular carcinoma Significant (P <0 05). (3) The positive rates of Gal 3 and MUC 1: benign lesions were 38 4% and 23 3% respectively, the positive rates of malignant tumors were 88 1% and 59 0% respectively. The positive rates of nodular goiter and adenoma ) Was lower than that of papillary thyroid carcinoma and follicular carcinoma (P <0.05). The expression of Gal 3 and MUC1 was basically parallel in thyroid carcinoma, but the sensitivity of Gal 3 was higher than that of MUC1. Gal 3 and MUC1 are both positive. While cytoplasmic TTF 1 expression is related to Gal 3, MUC1 positive expression. Conclusion TTF 1 nuclear reaction is normal
