目的探讨自发性高血压大鼠(SHR)和卒中易感型自发性高血压大鼠(SHRSP)肾脏组织中3-羟基-3-甲基戊二酰辅酶A合成酶2(HMGCS2)的表达差异及其在抗氧化中的作用。方法随机选取2月龄雄性SHR和SHRSP大鼠各50只,用免疫印迹法检测肾组织中HMGCS2和过氧化氢酶表达差异以及细胞信号磷酸化水平;同时测量组织中丙二醛(MDA)含量。结果SHRSP组HMGCS2表达量(6 398±658.128)明显低于SHR组(16516.67±1321.855)(P<0.05)。SHRSP组MDA含量[(0.738±0.175)U/mg prot]高于SHR组[(0.330±0.032)U/mg prot](P<0.05);SHRSP组蛋白激酶B(Akt)的磷酸化水平(1.909±0.124)明显高于SHR组(0.229±0.071)(P<0.05)。结论HMGCS2在SHRSP和SHR肾脏的表达差异有统计学意义,并可以在抗氧化中发挥作用;其表达可能由磷酸化蛋白激酶B(p-Akt)信号调节。
Objective To investigate the expression of 3-hydroxy-3-methylglutaryl coenzyme A synthetase 2 (HMGCS2) in spontaneously hypertensive rats (SHR) and spontaneous stroke-prone spontaneously hypertensive rats And its role in antioxidation. Methods 50 SHRSP and 2 male SHRSP rats were randomly selected. The expression of HMGCS2 and catalase in renal tissue and the phosphorylation of cell signal were detected by Western blotting. The contents of malondialdehyde (MDA) . Results The expression of HMGCS2 in SHRSP group (6 398 ± 658.128) was significantly lower than that in SHR group (16516.67 ± 1321.855) (P <0.05). The level of MDA in SHRSP group was significantly higher than that in SHR group ([(0.738 ± 0.175) U / mg prot] [(0.330 ± 0.032) U / mg prot] ± 0.124) was significantly higher than that of SHR group (0.229 ± 0.071) (P <0.05). Conclusion The expression of HMGCS2 in SHRSP and SHR kidneys is statistically significant and may play an important role in antioxidation. The expression of HMGCS2 may be regulated by phosphorylated protein kinase B (p-Akt) signaling.
