目的观察Glucan对刀豆蛋白A(ConA)所致小鼠免疫性肝损伤的干预效果及其作用机制。方法ConA尾静脉注射制备小鼠免疫性肝损伤模型,在ConA注射前1h腹腔注射Glucan,观察正常对照组、ConA损伤组、Glucan/ConA组肝损伤的情况,检测血清谷丙转氨酶(ALT)、α肿瘤坏死因子(TNF-α)及组织中一氧化氮(NO)含量的变化,并用DNAladder检测肝细胞凋亡的变化。结果ConA尾静脉注射后,出现明显的肝损伤,血清ALT、TNF-α及组织中NO水平明显升高,DNAladder检测可见明显的梯状条带;预先应用Glucan后,可减轻这些改变。结论Glu-can对ConA所致小鼠免疫性肝损伤有保护作用。
Objective To observe the effect of Glucan on immunological liver injury induced by ConA in mice and its mechanism. Methods The mouse model of autoimmune liver injury was established by tail vein injection of ConA. Glucan was intraperitoneally injected 1 hour before ConA injection to observe the liver injury in normal control group, ConA injury group and Glucan / ConA group. Serum alanine aminotransferase (ALT) α, tumor necrosis factor-α (TNF-α) and nitric oxide (NO) in the tissue were detected by flow cytometry. Results After the caudal vein injection of ConA, obvious hepatic injury occurred. The serum levels of ALT, TNF-α and NO in the tissues were significantly increased. DNA ladder showed obvious ladder-like bands. Pretreatment with Glucan alleviated these changes. Conclusion Glu-can can protect ConA-induced immune liver injury in mice.
