目的:初步探讨黄花倒水莲提取物对小鼠辐射损伤的防护作用。方法:以辐射损伤小鼠作为研究对象,分别以黄花倒水莲提取物2,4,8 g·kg-1·d-1灌胃,阴性对照组、辐射模型组给予等量生理盐水。灌胃5 d后,除阴性对照组外各组小鼠接受6 Gy的60Coγ射线一次性全身均匀照射,然后再继续灌胃10 d,最后1次灌胃24 h后,检测各组小鼠外周血中红细胞数(RBC)、白细胞数(WBC)、血小板数(PLT)、淋巴细胞数(Lymph),计算脾指数和胸腺指数,用单细胞凝胶电泳技术检测小鼠脾细胞、骨髓细胞的DNA损伤。结果:与阴性对照组比,辐射模型组、黄花倒水莲各剂量组小鼠的RBC数、WBC数、PLT数、Lymph数、胸腺指数、脾指数明显减少(P<0.01或P<0.05),脾细胞、骨髓细胞的尾部DNA百分率(Tail DNA%)和尾矩(Tail Moment)明显增大(P<0.01或P<0.05);但与辐射模型组比,黄花倒水莲各剂量组小鼠的RBC数、WBC数、PLT数、Lymph数、胸腺指数、脾指数明显增加(P<0.01或P<0.05),脾细胞、骨髓细胞的Tail DNA%和Tail Moment明显减少(P<0.01),存在剂量-反应关系。结论:黄花倒水莲提取物对辐射所致小鼠造血系统和免疫器官损伤有一定的防护作用。
Objective: To investigate the protective effect of the extract of Pseudostellaria dahurica against radiation injury in mice. Methods: Radiation-injured mice were used as research object. The rats in Radix et Rhizoma pallidum were treated with 2, 4, 8 g · kg-1 · d-1 gavage, negative control group and radiation model group respectively. After 5 days of intragastric administration, except for the negative control group, each group of mice received a single dose of 6 Gy of 60Co γ-rays and then continued to gavage for 10 days. After the last intragastric administration for 24 hours, The numbers of RBC, WBC, PLT and Lymph in blood were measured, splenic index and thymus index were calculated, and the spleen cells, the myeloid cells were detected by single cell gel electrophoresis DNA damage. Results: Compared with the negative control group, the numbers of RBCs, WBCs, PLTs, Lymphs, thymus index and spleen index in the radiation model group and the Danshuaxie lotus group were significantly decreased (P <0.01 or P <0.05) , Tail DNA% and Tail Moment in spleen cells and bone marrow cells increased significantly (P <0.01 or P <0.05). However, compared with radiation model group, The numbers of RBCs, WBCs, PLTs, Lymphs, thymus index and spleen index were significantly increased (P <0.01 or P <0.05), and the Tail DNA% and Tail Moment of spleen cells and bone marrow cells were significantly decreased , There is a dose-response relationship. CONCLUSION: The extract of Pseudostellaria Ligustris has some protective effects on the hematopoietic system and immune organ in mice induced by radiation.
