目的制备人原发性胆囊癌单克隆抗体,对其特异性进行研究。方法以胆囊癌细胞系SGC-996免疫Balb/c小鼠,应用淋巴细胞杂交瘤技术,获得稳定分泌抗SGC-996单克隆抗体的阳性杂交瘤细胞。结果杂交瘤细胞符合融合细胞核型特点,有较强的成瘤性。抗体亚类为IgGl。相对应的抗原主要表达在胞质和胞核中,分子量约为67kD。抗体对胆囊癌有较强的免疫反应,其他癌组织、癌旁组织及间质、正常胆囊上皮和成纤维细胞均呈阴性。结论建立一株杂交瘤细胞系,能稳定分泌单克隆抗体,陔抗体具有较高特异性。
Objective To prepare a monoclonal antibody against human primary gallbladder carcinoma and study its specificity. Methods Balb / c mice were immunized with the gallbladder carcinoma cell line SGC-996 and the hybridoma cells stably secreting anti-SGC-996 monoclonal antibody were obtained by lymphocyte hybridoma technique. Results hybridoma cells in line with the characteristics of the fusion cell karyotype, a strong tumorigenicity. The antibody subclass is IgG1. The corresponding antigens are mainly expressed in the cytoplasm and nucleus, with a molecular weight of about 67 kD. Antibodies have a strong immune response to gallbladder cancer, other cancer tissues, paracancerous tissues and stroma, normal gallbladder epithelium and fibroblasts were negative. Conclusion The establishment of a hybridoma cell line can stably secrete monoclonal antibodies, antibodies with high specificity.
